What Is 3,4-CTMP?

3,4-CTMP (3,4-dichloromethylphenidate) is a synthetic compound that is structurally related to methylphenidate. It has been identified by researchers as a novel psychoactive substance (NPS) and a methylphenidate analogue. The compound differs from methylphenidate only in terms of the presence of two chlorine atoms on the 3 and 4 positions of its phenyl ring.

The change in structure has prompted scientists to look at 3,4-CTMP in an effort to better understand how chemical modifications can impact the activity of dopamine and noradrenaline transporters.

3,4-CTMP R&D

Researchers first became interested in 3,4-CTMP while investigating compounds similar to methylphenidate and their pharmacological applications. But it never was developed into an approved drug. The compound was then marketed as a new psychoactive substance and drew scientific interest because of its strong effects on monoamine neurotransmitter systems.

Published research is still very limited compared to established medicines such as methylphenidate. Therefore, they cannot assume that effects observed in laboratory experiments can be safely or predictably translated to humans.

How does 3,4-CTMP work?

Dopamine Transporter Activity

Studies have shown that 3,4-CTMP acts on the dopamine transporter (DAT). The transporter typically removes dopamine from the space between nerve cells. 3,4-CTMP inhibits the reuptake of dopamine and thus increases the amount of dopamine in experimental models.

One in vitro study found that 3,4-CTMP caused a significantly greater increase in electrically stimulated dopamine efflux than methylphenidate over several tested concentrations. Much of this effect, researchers suggested, could be due to inhibition of DAT.

Noradrenaline actions

3,4-CTMP also modulated noradrenaline efflux in the experimental model. The same study showed that the experiment with the relevant brain region showed about a 2-fold increase at the highest concentration tested. But the results were on rat brain slices, not clinical human trials.

What the Scientific Evidence Shows

Another study in 2018 compared 3,4-CTMP with methylphenidate and ethylphenidate in rat brain-slice experiments, published in Frontiers in Psychiatry. Fast cyclic voltammetry was used to measure the release of dopamine and noradrenaline.

Under the experimental conditions, 3,4-CTMP was found to be a more potent stimulator of dopamine release than the comparison compounds. The researchers pointed out the possible addictive liability of the compound and called for caution in interpreting its pharmacological profile.

Importantly, these findings do not indicate a safe human dose, medical benefit, or approved therapeutic use of 3&4-CTMP.

Limitations of the Research and Safety

Absence of human evidence

The most significant issue with 3,4-CTMP is the lack of full human safety data. The pharmacology of 3&4-CTMP has been studied in laboratory models, but that evidence cannot be used to establish long-term safety in people.

Possibility of Addiction

Published studies on 3&4-CTMP have raised concerns about the possibility of addictive or dependence-related effects due to its strong effects on dopamine signalling in experimental systems.

Product Purity May Vary

Research chemicals sold outside of controlled scientific environments may be of unknown identity, concentration or purity. Analytical testing is thus important in real laboratory research.

A Study of 3,4-CTMP

The evaluation of 3,4-CTMP may include gas chromatography-mass spectrometry (GC-MS), nuclear magnetic resonance (NMR) and liquid chromatography-mass spectrometry. It has also been investigated for its metabolites by high-resolution mass spectrometry. In one metabolite study several phase-I metabolites were identified, and it was concluded that hydrolysis plays an important role in its metabolism.

But scientists know little about its effects on humans and if it is safe to use long term.

Therefore, 3,4-CTMP is neither an approved drug nor a dietary supplement. Scientific studies still provide the right basis for understanding chemistry, pharmacology, metabolism and potential risks.
